طريقة عمل الفحص البكتيري بالتفصيل  :

 Methods of Bacteriological Examination in Detail

1- يجب أن تكون جميع المعدات والزجاجيات معقمة وجاهزة .

                                             All equipment and glassware must be sterile and ready.

2- هذا هو الـ glass funnel  الذي يتم به تجميع الــ sample  ، يجب أن يكون معقم بالأوتوكلاف .

     This is the glass funnel where the sample is collected, it must be autoclave  sterilized.

      

3- هذا هو الــ clamp  ليمسك الــ funnel  بقاعدة الترشيح .

                                                           This is the clamp to hold the funnel with the filter base.

     

4- قاعدة الترشيح تكون موصولة بـ Vacuum pump حيث يسحب أو يشفط وعمل    Vacuum ليسحب الـ Sample  من أعلى قاعدة الترشيح .

                   The filter base is connected to a vacuum pump where it is pulled or sucked and 

                                          worked  Vacuum to pull the sample from the top of the filter base.

     

5- يجب ان يكون هناك لهب بنسن  Bunsen  لتعقيم الوسط المحيط بعملية الفحص البكتيري        

    وتعقيم بعض الزجاجيات المستخدمة في الفحص البكتيري بتعريضها للحرارة . 

      There should be a Bunsen flame to sterilize the surrounding medium with the bacterial   screening process and sterilize some of the glass used in the bacterial examination by exposing it to heat.

    

6- نقوم بإشعال لهب بنسن ضروري أن يكون في العمل حتى يعقم الوسط المحيط .

   We ignite the flame of the benson necessary to be in action until the surrounding medium  is sterilized.

    

7- نعرض الـ funnel  وقاعدة الترشيح إلى لهب بنسن لتعقيمه

                                 We offer the funnel and filtration base to the Bensen flame to sterilize it

8- نعقم الملقط بواسطة الإيثانول وكذلك تعريضه للهب

     

                                            Sterilize the forceps by ethanol as well as expose them to flame

     

9- نقوم بإخراج ورقة الترشيح Cellulose Nitrate Filter  مساميته 0.45 ميللي ماكرون
     يجب أن تكون العبوات محكمة الإغلاق .

       We remove the filter paper Cellulose Nitrate Filter 0.45 MILLY MACRON The packages   must be sealed.
  

   

 ملاحظة : 

عند نزع ورقة الترشيح عن الغطاء الذي يغلفها يتم ذلك من خلال الملقط المعقم                     

 واحذر ملامسة  يدك أو إصبعك لورقة الترشيح .
Note :

When removing the filter paper from the cover that is wrapped, do this with sterile forceps and be careful not to touch your hand or finger with the filter paper.

10 – نسحب ورقة الترشيح بواسطة الملقط ونضعها على قاعدة الترشيح الزجاجية .

                                 Pull the filter paper with the forceps and place it on the glass filter base.
      

11- نأتي بالــ glass funnel ونضعه فوق قاعدة الترشيح .

                                                          Bring the glass funnel and place it above the filter base.

      

12- نمسك ونثبت الــ glass funnel بقاعدة الترشيح الزجاجية بواسطة الــ Clamp  والوضع جاهز لإضافة عينة الماء المراد فحصها .

        Clamp and fix the glass funnel with the glass filter base by Clamp and ready to add  the water sample to be tested.

    

13- نأتي بطبق التحضين الجاهز الذي تكون به الميديا جاهزة في داخل الطبق ويجب أن يكون علبة الأطباق معقمة ومغلقة بإحكام حتى لا يحدث له أي تلوث بكتيري .

We come with a ready-made dish, in which the media is ready inside the dish, and the tray of dishes must be sterile and tightly closed so that it does not cause any bacterial contamination.

    

14- نأخذ 3 مل ماء مقطر بواسطة الماصة وتكون للاستخدام مرة واحدة أو معقمه لوكانت زجاجية ونرطب طبق التحضين بالماء المقطر حتى يصبح الطبق جاهز لاستقبال عينة الفحص البكتيري .

Take 3 ml of distilled water with the pipette and be used once or sterilized if it is glass and moisturize the dish with the distilled water until the dish is ready to receive the sample of the bacterial examination.

    

  

15- نجمع العينة المراد الفحص البكتيري لها بواسطة glass  خاص يكون محكم الإغلاق ومعقم سابقا بالأوتوكليف حيث نضيف عينة الماء داخل الــ glass funnel   لغاية 100 مل فقط .

We collect the sample for bacterial examination using a special glass that is tightly sealed and autoclave. We add the water sample in the glass funnel to 100 ml only.

     

16- نشغل الفاكيوم vacuum pump  لسحب عينة الماء اللازم للــ الفحص البكتيري حيث يسحب عينة الماء للأسفل ويكون قد نفذ الماء من ورقة الترشيح التي لا تسمح بنفاذ  البكتيريا وإمساكها على سطحها 

The vacuum pump is used to extract the sample of water required for the bacterial examination. The water sample is drawn down and the water is applied from the filter paper, which does not allow bacteria to enter and hold on the surface.

   

17- نمسك ورقة الترشيح بواسطة ملقط معقم ونضعها في داخل طبق الفحص البكتيري 

                 Press the filter paper with sterile forceps and place it inside the bacterial test dish
    

    

18- نضع طبق الباتري في جهاز الإنكيوبيتر Incubator  الحاضنة ونضبطه عند درجة حرارة 38 Cلأن هذا النوع من البكتيريا تنمو عند هذه الدرجة من الحرارة 36 C   (+ أو – 2 )

 Place the patri dish in the Incubator incubator and adjust it at 38 ° C because this type of bacteria grows at this temperature of 36 C (+ or - 2)
   

19 – بكتيريا الإيكولاي E.coli  تبدو على الطبق بلون نقاط حمراء ذات لمعان معدني محمر .

                                           E.coli appears on the dish with red dots of reddish metallic gloss.
     
     

20 – بكتيريا الكوليفورم Coliform تكون على شكل نقاط حمراء فاتحة .

                                                                    Coliform bacteria are in the form of light red dots.
